James Bond, agent des services spéciaux britanniques, est envoyé en mission à la Jamaïque où le correspondant anglais et sa secrétaire ont été exécutés par une mystérieuse organisation. Il fait la connaissance de son homologue américain, Leiter, agent du FBI, qui enquête sur les activités du docteur No, un asiatique propriétaire d'une petite île des environs, Crab Key, sur laquelle il est interdit d'aborder.
